Maintenance of the information embedded in the genomic DNA sequence is essential for life. DNA polymerases play pivotal roles in the complex processes that maintain genetic integrity. Besides their tasks in vivo, DNA polymerases are the workhorses in numerous biotechnology applications such as the polymerase chain reaction (PCR), cDNA cloning, genome sequencing, nucleic acids-based diagnostics and in techniques to analyze ancient and otherwise damaged DNA. Moreover, some diseases are related to DNA polymerase defects, and chemotherapy through inhibition of DNA polymerases is used to fight HIV, Herpes and Hepatitis B and C infections. We have recently witnessed the discovery of an abundance of novel DNA polymerases in viruses, bacteria, archaea and eukaryotes with specialized properties whose physiological functions are only beginning to be understood. This book summarizes the current knowledge of these fascinating enzymes. It is intended for a wide audience from basic scientists, to diagnostic laboratories and to clinicians who seek a better understanding of these fascinating enzymes.

The Metaphysical Vision: Arthur Schopenhauer's Philosophy of Art and Life and Samuel Beckett's Own Way to Make Use of It expands upon the ideas and theories set forth in the author's Die eigentlich metaphysische Tätigkeit: Über Schopenhauers Ästhetik und ihre Anwendung durch Samuel Beckett, published (in German) in 1982 and hailed by Catharina Wulf in her book The Imperative of Narration (1997) as an «excellent study» and «the most thorough enquiry into Beckett and Schopenhauer.» In the last years of the twentieth century, new documents regarding Samuel Beckett's reading and thinking, especially important notebooks and letters, have become accessible to scholars. These documents show much more clearly than could ever be demonstrated previously that Beckett had a strong, lifelong interest in Schopenhauer's philosophy. There is no other philosopher to whom Beckett refers more often in his personal comments throughout the years of his writing up to his seventies; no other philosopher whose view of life and the world comes closer to the image of human existence we find in Samuel Beckett's literary work. The striking similarity in matters of world view and human life, and especially the evidence obtained from Beckett's previously unknown notebooks and letters, call for a close systematic study of the Beckett-Schopenhauer relationship. Ulrich Weisstein, an international authority in the fields of comparative literature and comparative arts, has been a pioneer paving the way for present-day intermedia studies. Among his broad intermedial interests opera has always held a central place. For the first time this volume makes available his major contributions to opera criticism in compact form, thus meeting a serious scholarly demand. The necessarily stringent selection of essays from Professor Weisstein's large output on opera, reflecting fifty years of involvement with the genre, is primarily governed by the wish to present texts that are representative of their author's work and, at the same time, are unlikely to be readily available through other channels. Premodern societies believed in something sacred that obliged unconditionally. Modern societies rely on fallible science. Do they also need something absolute, a secular sacred? Steinvorth analyzes the writings of modern philosophers who claim that there is an absolute norm: the norm to be rational and authentic. In his view, their claim is true if it is reinterpreted. The norm is not moral, as it was thought to be, but metaphysical, and authenticity is not self-realization, but doing things for their own sake. In discussing the pros and cons of philosophical claims on absolutes, this book spreads out the rich pool of philosophical ideas and clarifies urgent contemporary questions about what can be demanded with universal validity. It argues this is not only the principle of justice, not to harm, but also a metaphysical principle by which to find meaning in life. Moreover, it points to some consequences this principle has in politics.
