Excitable Cells in Tissue Culture

The tissue culture approach to the study of membrane properties of excitable cells has progressed beyond the technical problems of culture methodology. Recent developments have fostered substantive contributions in research con cerned with the physiology, pharmacology, and biophysics of cell membranes in tissue culture. The scope of this volume is related to the application of tissue culture methodology to developmental processes and cellular mechanisms of electrical and chemical excitability. The major emphasis will be on the body of new biological information made available by the analytic possibilities inherent in the tissue culture systems. Naturally occurring preparations of excitable cells are frequently of suf ficient morphological complexity to compromise the analysis of the data obtained from them. Some of the limitations associated with dissected prepa rations have to do with the direct visualization of and access to the cell(s) in question and maintenance of steady-state conditions for prolonged periods of time. Since preparations in tissue culture can circumvent these problems, it is feasible to analyze the properties of identifiable cells, grown either singly or in prescribed geometries, as well as to follow the development of cellular inter actions. A crucial consideration in the use of cultured preparations is that they must faithfully capture the phenomenon of interest to the investigator. This and other potential limitations on the methodology are of necessary concern in the present volume.

There is growing pressure and stress placed on organisations to fight for customers and service/product placement in an increasingly competitive global marketplace. It has, therefore, never been more important to get the best out of the workforce. To achieve this, the role of the leader can be a fundamental factor in organisational success or failure. Leaders need to have the requisite skills to reflect the demands placed upon them in the 21st century. There are the “accidental managers” who just drop into the role of leadership and others who may develop skills and knowledge in readiness for a leadership role. There are also those who may have the innate ability to lead. Within the mix are those who are characterised by traits associated with the “dark triad” or who may use “pathocratic influence” on others to conform, reinforcing values (or lack of values) associated with toxic leadership. They create damage and harm. They become “passion killers”. The result can lead to a “pathocracy”. This book discusses the role emotional intelligence plays in helping people deal with stressful and challenging experiences, suggesting different ways to cope. The author reflects on the values that are integral to the success or failure of an organisation. “Passion” is identified as an added value that can differentiate one organisation from another. If passion is harmed, it can affect motivation, creativity, output, performance, and productivity. Therefore, this book provides the reader with examples of “passion killing” while making suggestions as to factors that can be adopted to engage and encourage passion. Conclusions are drawn and recommendations made to support those faced with “passion killers”. This book is aimed at those of all ages and educational backgrounds interested in developing their leadership knowledge and skills. It is also aimed at those interested in learning more about differences in personality, emotional intelligence, stress, coping, values, and the importance of understanding the impact of “passion killers”.

An insider’s account of a wrongful conviction and the fight to overturn it during the civil rights era This book is an insider’s account of the case of Freddie Pitts and Wilbert Lee, two Black men who were wrongfully charged and convicted of the murder of two white gas station attendants in Port St. Joe, Florida, in 1963, and sentenced to death. Phillip Hubbart, a defense lawyer for Pitts and Lee for more than 10 years, examines the crime, the trial, and the appeals with both a keen legal perspective and an awareness of the endemic racism that pervaded the case and obstructed justice. Hubbart discusses how the case against Pitts and Lee was based entirely on confessions obtained from the defendants and an alleged “eyewitness” through prolonged, violent interrogations and how local authorities repeatedly rejected later evidence pointing to the real killer, a white man well known to the Port St. Joe police. The book follows the case’s tortuous route through the Florida courts to the defendants’ eventual exoneration in 1975 by the Florida governor and cabinet. From Death Row to Freedom is a thorough chronicle of deep prejudice in the courts and brutality at the hands of police during the civil rights era of the 1960s. Hubbart argues that the Pitts-Lee case is a piece of American history that must be remembered, along with other similar incidents, in order for the country to make any progress toward racial reconciliation today. Contemporary architecture, and the culture it reflects dependent as it is on fossil fuels, has contributed to the cause and necessity of a burgeoning green process that emerged over the past half century. This text is the first to offer a comprehensive critical history and analysis of the greening of architecture through accumulative reduction of negative environmental effects caused by buildings, urban designs and settlements. Describing the progressive development of green architecture from 1960 to 2010, it illustrates how it is ever evolving and ameliorated through alterations in form, technology, materials and use and it examines different places worldwide that represent a diversity of cultural and climatic contexts. The book is divided into seven chapters: with an overview of the environmental issues and the nature of green architecture in response to them, followed by an historic perspective of the pioneering evolution of green technology and architectural integration over the past five decades, and finally, providing the intransigent and culturally pervasive current examples within a wide range of geographic territories. The greening of architecture is seen as an evolutionary process that is informed by significant world events, climate change, environmental theories, movements in architecture, technological innovations, and seminal works in architecture and planning throughout each decade over the past fifty years. This time period is bounded on one end by the awareness of environmental problems beginning in the 1960's, the influential texts by Rachel Carson, E.F. Schumacher, Buckminster Fuller and Steward Brand, and the impact of the OPEC Oil Embargo of 1973, and on the other end the pervasiveness of the necessary greening of architecture that includes, systemic reforms in architectural and urban design, land use planning, transportation, agriculture, and energy production found in the 2000's. The greening process moves from remediation to holistic models of architecture. Geographical landscapes give a global account of the greening process where some examples are parallel and sympathetic, and others are in clear contrast to one another with very individuated approaches. Certain events, like the Rio Summit in 1992 and Kyoto Protocol in 1997, and themes, such as the Hannover Principles in 2000, provide a dynamic ideological critique as well as a formal and technical discussion of the embodied and accumulative content of greening principles in architecture.

The overview is eclectic; it presents views commonly held by instructional designers that have been synthesized from the research and literature. This book differs from most books on instructional design in that it describes a process that is internally consistent; that is, it embraces a specific approach which makes it possible and easier for the designer to achieve closure on a process. Most books on instructional design describe several processes without adequately providing an opportunity for closure. As a result, many designers go away confused, owning bits and pieces of a process that it is impossible, or at least difficult, to implement in a coherent fashion.
