Lake Ronkonkoma, located in the center of Long Island, became a summer destination for wealthy New Yorkers with the arrival of the railroad in 1843. Luxurious hotels were built around the lakeshore during the 1890s to attract visitors. People would utilize the sparkling waters of the lake for boating, fishing, and swimming. At night, hotel guests could dance, gamble in the casino, and enjoy sumptuous meals. By the 1920s, as more people could afford automobiles, beach pavilions began decorating the shoreline of Lake Ronkonkoma. Thousands of people were flocking to this summer paradise. Canoeing, camping, dancing to live music, beauty contests, and fireworks entertained visitors. The area continued to grow in popularity until World War II when gas rationing limited travel. By the 1950s, Lake Ronkonkoma had become a town populated by year-round residents.

The motivation for us to conceive this series of volumes on regulation was mainly our belief that it would be fun, and at the same time productive, to approach the subject in a way that differs from that of other treatises. We thought it might be interesting and instructive for both author and reader-to examine a particular area of investigation in a framework of many different problems. Cutting across the traditional boundaries that have separated the subjects in past volumes on regulation is not an easy thing to do-not because it is difficult to think of what interesting topics should replace the old ones, but because it is difficult to find authors who are willing to write about areas outside those pursued in their own laboratories. Anyone who takes on the task of reviewing a broad area of interest must weave together its various parts by picking up the threads from many different laboratories, and attempt to produce a fabric with a meaningful design. Finding persons who are likely to succeed in such a task was the most difficult part of our job. In the first volume of this treatise, most of the chapters dealt with the mechanisms of The second volume involved a somewhat regulation of gene expression in microorganisms. broader area, spanning the prokaryotic-eukaryotic border. Topics ranged from phage mor phogenesis to the role of gradients in development. The last volume-Volume 3A-con cerned hormones, as does this volume-Volume 3B.

Since the 1930s and the work of Hale and Warkany on birth defects produced by variations in dietary vitamin A, it has been recognised that the developing fetus is particularly vulnerable to adverse influences in the environment. . Studies of malformations at birth remained largely in the hands of paediatricians who, for instance, quickly established the con nection between rubella infection in early pregnancy and the birth of severely affected infants. However, it was through the tragic events of 1962, when dramatic increases in the incidence of phocomelia in newborn infants in Germany, the United Kingdom, Japan, and other countries were traced to the use of the apparently non-toxic sedative thalidomide by pregnant women, that toxicologists were brought face to face with the devastating possibility of chemically-induced developmental abnormalities. It had been shown earlier that chemotherapeutic agents could cause damage to the developing organism, but the absence of any known examples of drug-induced birth defects had produced an air of complacency which was reinforced by the lack of any specific regulatory requirements for safety evaluation in this area. The magnitude of the thalidomide tragedy, affecting some 10000 malformed children, was sufficient to cause an immediate, and some would say hasty, reaction by government drug regulatory agencies throughout the world to construct a test protocol which would detect potential teratogenic substances that might give rise to anatomical malformations.
