Scientists are deciphering the biology of the tumor cell at a level of detail that would have been hard to imagine just a decade or so ago. The development of high-throughput DNA sequencing and genomics technologies have allowed an understanding of the development, growth, survival, and spread of cancer cells in the body. From this information, we now have a basic blueprint or roadmap of how a single damaged cell can develop into a pre-malignant lesion, a primary tumor, and finally, a lethal tumor that may spread throughout the body and resist both medical therapy and host immune responses. In this book, we provide an overview of our current understanding of this cancer blueprint, which has been aided both by the study of familial cancer syndromes, in vitro studies of cancer cells, and animal models. Three classes of genes have emerged from these studies: tumor suppressor genes needed for normal growth control and DNA repair; oncogenes that regulate cell growth and survival, and epigenetic modifiers, enzymes that regulate the modification of DNA and the proteins that form chromatin. Each of these three classes of genes is mutated or altered at least once in virtually all malignant cancer cells. Current technologies permit the DNA sequencing of cancer exomes (coding gene sequencing), whole genomes, transcriptome (all expressed genes), and DNA methylation profiling. These studies show that all tumors have unique constellations of mutated, rearranged, amplified, and deleted genes. Single-cell sequencing further shows that there is extensive variation in individual cells in the tumor; that cancers evolve, and have many of the properties of a multi-cellular entity. Lastly, cancer cells, through mutations in epigenetic modifiers, can reprogram the genome and unlock entire developmental and gene expression pathways to adapt and survive in changing conditions. This reprogramming allows the tumor to elude the host body's defenses, radiotherapy, chemotherapy, and targeted therapy that we use in cancer treatment. Understanding this cancer blueprint paves the way for the development of future therapies to treat and eliminate cancer.

Although Jews in early modern Germany produced little in the way of formal historiography, Jews nevertheless engaged the past for many reasons and in various and surprising ways. They narrated the past in order to enforce order, empower authority, and record the traditions of their communities. In this way, Jews created community structure and projected that structure into the future. But Jews also used the past as a means to contest the marginalization threatened by broader developments in the Christian society in which they lived. As the Reformation threw into relief serious questions about authority and tradition and as Jews continued to suffer from anti-Jewish mentality and politics, narration of the past allowed Jews to re-inscribe themselves in history and contemporary society. Drawing on a wide range of sources, including chronicles, liturgical works, books of customs, memorybooks, biblical commentaries, rabbinic responsa and community ledgers, this study offers a timely reassessment of Jewish community and identity during a frequently turbulent era. It engages, but then redirects, important discussions by historians regarding the nature of time and the construction and role of history and memory in pre-modern Europe and pre-modern Jewish civilization. It expounds in detail the characteristics of quasiverbals, verbless clauses, discontinuous and double-duty clausal constituents, and supra-clausal structures. The book is intended for students of Biblical Hebrew at all levels. A crisis is coming for everyone who uses math and science. For decades now, the classical model of probability (the indifference principle and the Gaussian distribution) has been breaking down and revealing its limitations in fields from economics to epidemiology. Now a new approach has revealed the underlying non-classical principle behind all these 'anomalous' laws: — Pareto’s law of elite incomes — Zipf’s law of word frequencies — Lotka’s law of scientific publications — Kleiber’s law of metabolic rates — the Clausewitz-Dupuy law of combat friction — Moore’s law of computing costs — the Wright-Henderson cost law — Weibull’s law of electronics failures — the Flynn Effect in IQ scores — Benford’s law of digit frequencies — Farr’s law of epidemics — Hubbell’s neutral theory of biodiversity — Rogers’ law of innovation classes — Wilson’s law of island biogeography — Smeed’s law of traffic fatalities The general law behind all these particular laws (and countless others) is the "decline effect". As a system ages or grows in size, the rules of probability subtly change. Entropy increases, rare items become rarer, and average performance measures decline. The human meaning of a decline may be positive (decreasing costs, falling epidemic mortality) or negative (lower customer loyalty, decreasing efficiency), but the mathematical pattern is always the same. The implications are enormous, as these examples show: All epidemic diseases decline in infectiousness and in lethality. HIV-AIDS went from a highly infectious, 95-percent fatal disease, to a survivable condition with a latency of decades. COVID-19 went from a death rate of 7 percent in early 2020, to under 2 percent in 2022. Hereditary dynasties around the world declined smoothly in lifespan, from hundreds of years to tens of years. When democracies replaced monarchies, the decline (in spans of party control) continued.
