Tumourigenesis is the formation of tumours in the body, often caused by oncogenes. These tumours are the result of uncontrollable reproduction (cell division) due to alterations in the cell's genetic code, creating lesions in the tissue where they reside. Gold has traditionally been regarded as inactive as a catalytic metal. However, the advent of nanoparticulate gold on high surface area oxide supports has demonstrated its high catalytic activity in many chemical reactions. Why do we grow up to look, act, and feel as we do? Through most of the twentieth century, scientists and laypeople answered this question by referring to two factors alone: our experiences and our genes. But recent discoveries about how genes work have revealed a new way to understand the developmental origins of our characteristics. These discoveries have emerged from the new science of behavioral epigenetics--and just as the whole world has now heard of DNA, "epigenetics" will be a household word in the near future. Behavioral epigenetics is important because it explains how our experiences get under our skin and influence the activity of our genes. Because of breakthroughs in this field, we now know that the genes we're born with don't determine if we'll end up easily stressed, likely to fall ill with cancer, or possessed of a powerful intellect. Instead, what matters is what our genes do. And because research in behavioral epigenetics has shown that our experiences influence how our genes function, this work has changed how scientists think about nature, nurture, and human development. Diets, environmental toxins, parenting styles, and other environmental factors all influence genetic activity through epigenetic mechanisms; this discovery has the potential to alter how doctors treat diseases, and to change how mental health professionals treat conditions from schizophrenia to post-traumatic stress disorder. These advances could also force a reworking of the theory of evolution that dominated twentieth-century biology, and even change how we think about human nature itself. In spite of the importance of this research, behavioral epigenetics is still relatively unknown to non-biologists. Nuclear receptors are transcription factors that bind steroid, retinoid and thyroid hormones, and other ligands to drive hormone-dependent gene expression in conjunction with co-activators and co-repressors, collectively referred to as co-regulators. So far, more than 400 co-regulators have been reported in the literature and they have been implicated in a wide variety of pathological conditions, genetic syndromes, and in cancer. A key feature of co-regulator-based disease is the pleiotropic effects that disruption of normal co-regulator function has on energy metabolism, neurological function, and susceptibility to cancer. Technological advances in proteomics, genomics, and transcriptomics are leading to new ways to understand the pleiotropic actions of co-regulators. We expect that co-regulator ‘omics’ will lead to ways of understanding how co-regulators can be evaluated in the context of other complex genetic factors, hormones, diet, the environment, and stress. The broad role that co-regulators have in human pathological conditions makes it important to consider them as important new drug targets, such as for the treatment of hormone-dependent cancers or for indications related to energy metabolism. Better system-wide knowledge of co-regulator control of transcription and physiology is expected to lead to the best placement for future co-regulator-based therapies.
