The short period since the publication of Volume 1 of Methods in Membrane Biology has been a time of momentous progress. Calorimetry, electron spin and nuclear magnetic resonance, X-ray diffraction, and freeze-cleavage electron microscopy, reinforced by biochemical analyses and enzymatic studies, have led to universal acceptance of a generalized membrane model. All membrane biologists would agree that a major element of all biological membranes is a bilayer of phospholipids which, in some instances, also contains other lipids, notably sterols and glycolipids. The fatty acid com position of the lipids of most membranes is such that the lipids are above their transition temperatures in their normal environment so that the bilayer is fluid. The microviscosity of the fatty acyl groups decreases pro gressively down the chain so that, at the hydrocarbon interior of the bilayer, the lipid phase has a viscosity approximating that of olive oil at room temperature. As a consequence of this membrane fluidity, a phospholipid molecule is very mobile within the plane of the membrane (moving a distance of about 1-2 fLm in 1 s) but the movement of a phospholipid molecule from one side of the membrane bilayer to the other (flip-flop) is very slow. The lipid bilayer is an essentially inert and rather impermeable struc ture, as shown by many studies with model systems. Proteins, of course, provide the catalytic components of the membranes. as well as playing a significant structural role.
The contributions of electron microscopy to membrane biology have been indispensable and, at the same time, disappointing. Membranes were known to exist before the advent of electron microscopy and general principles of their composition and molecular organization had been deduced from permeability and electrical conductivity measurements, polarized light microscopy, and X-ray diffraction. On the other hand, the complexities of the many intracellular membranes and membranous organelles were really not suspected until they were observed by the electron microscopist. One then had further hopes that the high resolution of the electron microscope (theoretically it can resolve atomic distances) would allow the visualization of the molecular architecture of membranes and lead directly to an under standing of structure and function at the molecular level. This expectation has been largely unrealized. Because of the great difficulties encountered in the preparation of biological material, because of the uncertainty of the chemistry of "staining," and because of numerous electron optical artifacts, it has been a major challenge just to rationalize the observed images in terms of the known facts, let alone to utilize the images to expand our knowledge of the molecular structure of biological membranes. The many differences among membranes with respect to function and composition are lost in the universal trilamellar image. Perhaps the one major exception to this, and the major structural contribution of electron microscopy at the molecular level, has come from freeze-etch electron microscopy.
The field of DNA repair is vast and advancing rapidly. Recent investigations have begun to focus on the involvement of chromatin in the repair of broken DNA. Although I have no doubt that many breakthroughs in our understanding of chromatin, chromatin regulation, and DNA repair lie in our future, presently this is a new line in inquiry. As such there are many, many unanswered questions. Indeed, most of the correct questions have probably not even been asked yet. Here I have attempted to present a review of some of the current body of knowledge that may prove relevant to understanding the role of chromatin in DNA repair. Because the volume of research, and the relevant findings, come from a staggering array of labs, systems, and ideas I have focused primarily on findings developed from the study of the budding yeast Saccharomyces cerevisiae. Unfortunately, this means that I have left out a great deal of information. It is my hope, however, that the information I do detail, particularly in Chapter 1, will give a flavor for the scope of the problem and perhaps highlight some of the interesting directions this field is taking, or may one day take. I would also point out that the primary research that is presented herein is not in any way meant to represent the comprehensive scope of research being performed. To understand DNA repair will require investigation from innumerable labs, performed by innumerable researchers, moving in unexpected directions.

Recently there has been increased interest in the development of computer-aided design programs to support the system level designer of integrated circuits more actively. Such design tools hold the promise of raising the level of abstraction at which an integrated circuit is designed, thus releasing the current designers from many of the details of logic and circuit level design. The promise further suggests that a whole new group of designers in neighboring engineering and science disciplines, with far less understanding of integrated circuit design, will also be able to increase their productivity and the functionality of the systems they design. This promise has been made repeatedly as each new higher level of computer-aided design tool is introduced and has repeatedly fallen short of fulfillment. Sponsored by the Wright-Ingraham Institute, the conference brought together a wide range of scientists, humanists, public officials, representatives of industrial and agricultural organizations, and interested citizens concerned not only about this important regional problem, but about the broader implications of competing land-use needs. Conference participants examined factors important to changes in land use, giving particular attention to the natural, economic, political, and value systems at work on the watershed in terms of how these systems affect and will be affected by changing land-use patterns.
In Contesting Commemoration: The 1876 Centennial, Independence Day, and the Reconstruction-Era South, Jack Noe examines identity and nationalism in the post–Civil War South through the lens of commemorative activity, namely Independence Day celebrations and the Centennial of 1876. Both events presented opportunities for whites, Blacks, northerners, and southerners to reflect on their identity as Americans. The often colorful and engaging discourse surrounding these observances provides a fascinating portrait of this fractured moment in the development of American nationalism.
Lords of the Sea revises our understanding of the epic political, economic, and cultural transformations of Japan’s late medieval period (ca. 1300–1600) by shifting the conventional land-based analytical framework to one centered on the perspectives of seafarers who, though usually dismissed as "pirates," thought of themselves as sea lords. Over the course of these centuries, Japan’s sea lords became maritime magnates who wielded increasing amounts of political and economic authority by developing autonomous maritime domains that operated outside the auspices of state authority. They played key roles in the operation of networks linking Japan to the rest of the world, and their protection businesses, shipping organizations, and sea tenure practices spread their influence across the waves to the continent, shaping commercial and diplomatic relations with Korea and China. Japan's land-based authorities during this time not only came to accept the autonomy of "pirates" but also competed to sponsor sea-lord bands who could administer littoral estates, fight sea battles, protect shipping, and carry trade. In turn, prominent sea-lord families expanded their dominion by shifting their locus of service among several patrons and by appropriating land-based rhetorics of lordship, which forced authorities to recognize them as legitimate lords over sea-based domains. By the end of the late medieval period, the ambitions, tactics, and technologies of sea-lord mercenary bands proved integral to the naval dimensions of Japan’s sixteenth-century military revolution. Sea lords translated their late medieval autonomy into positions of influence in early modern Japan and helped make control of the seas part of the ideological foundations of the state.
